DSIP is a nonapeptide that occurs naturally. The sequence is Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu.
Researchers isolated it in 1977, from the cerebral venous blood of a rabbit.
02Mechanism: How It Works
The mechanism is poorly defined. No isolated receptor exists. No gene exists.
The proposal describes a neuromodulator of slow-wave sleep, of neurotransmitters, and of circadian and stress responses.
Reliability flag. Indexed reviews call the sleep-factor hypothesis weak.

Evidence tier: clinical, small and mixed.
Positive result. Intravenous DSIP at 25 nmol/kg improved sleep in people with chronic insomnia (Schneider-Helmert). PMID 7028502.
Negative results. A double-blind study found no major therapeutic benefit. PMID 1299794. A crossover study found effects that did not differ significantly from placebo. PMID 3583493.
Evidence limit. The mechanism is contested and the trial record does not settle it.
